چکیده

While we read with great interest the recent report concerning the reversal of dabigatran-induced coagulopathy [1], we are concerned that methodological issues limit the validity of the authors’ conclusions. Firstly, it is unclear why a porcine model was utilized. Although there is clinical relevance for testing reversal agents in a trauma model, all agents were administered ex vivo. Furthermore, the established correlation between the activated partial thromboplastin time (aPTT) and dabigatran concentrations in humans [2] is absent in the pig model. Potentially, porcine coagulation pa-

Prothrombin complex concentrates and a specific antidote to dabigatran are effective ex-vivo in reversing the effects of dabigatran in an anticoagulation/liver trauma experimental model

INTRODUCTION New oral anticoagulants are effective alternatives to warfarin. However, no specific reversal agents are available for life-threatening bleeding or emergency surgery. Using a porcine model of trauma, this study assessed the ability of prothrombin complex concentrate (PCC), activated PCC (aPCC), recombinant FVIIa (rFVIIa) and a specific antidote to dabigatran (aDabi-Fab) to reverse ...

The action of pepsin on porcine immunoglobulin M and its effect on biological activity.

Treatment of porcine immunoglobulin M (IgM) with pepsin at pH 4.6 and 37 degrees C was found to gradually remove Fab arms and Cmicro2 domains over a period of 18h. Structural studies failed to find any other change. The main products can therefore be regarded as IgM-like molecules with limited numbers of Fab arms and Cmicro2 domains. Results indicated that this removal of Fab arms is probably a...

Evaluation of surrogate markers for human immunodeficiency virus infection among blood donors at the blood bank of "Hospital Universitário Regional Norte do Paraná", Londrina, PR, Brazil.

This study evaluated the usefulness of the anti-HBc, hepatitis C virus antibodies (anti-HCV), human T cell lymphotropic virus I and II antibodies (anti-HTLV I/II), serologic tests for syphilis, and surface antigen of hepatitis B virus (HBsAg) as surrogate markers for the risk for HIV infection in 80,284 serum samples from blood donors from the Blood Bank of "Hospital Universitário Regional Nort...

Monoclonal Antibodies Against Porcine Platelet Membrane Glycoproteins lb and

We prepared murine monoclonal antibodies to porcine platelet membrane glycoprotein (GP) lb and GP llb/Illa for further study of the porcine hemostatic mechanism. One monoclonal antibody, designated PP3-4C. blocked Ristocetin-induced platelet agglutination and caused 80% inhibition of Ristocetin-induced ‘25l-von Willebrand factor (vWF) binding to porcine platelets at a concentration of 1 2 gsg l...

Construction of a human recombinant polyclonal Fab fragment antibody library using peripheral blood lymphocytes of snake bitten victims

Human snake bitten poisoning is a serious threat in many tropical and subtropical countries such as Iran. The best acceptable treatment of envenomated humans is antivenoms; however they have a series of economic and medical problems and need more improvements. In this study a combinatorial human immunoglobulin gene library against some of Iranian snakes venoms was constructed. Total RNA prepare...
